چکیده

cientists in the field of blood cell development have found a protein of expansive abilities. According to two new papers, HoxB4 is able to promote self-renewal of hematopoietic stem cells (HSCs) both in vivo and in vitro. HoxB4 is a homeotic gene whose expression in vivo has been shown previously to enhance HSC regeneration. Now, Jennifer Antonchuk, Guy Sauvageau, and Keith Humphries (Terry Fox Laboratory, Vancouver, BC) demonstrate that the number of adult HSC can be expanded in vitro when bone marrow cells are transduced with HoxB4 . Compared with untransfected cells, the HoxB4expressing cells showed a 40-fold net increase after 14 d in culture. “So far, this [method] stands out uniquely as a means to expand stem cells,” says Humphries. “The stem cells are normal in terms of being able to S

Expansion of Non-Enriched Cord Blood Stem/Progenitor Cells CD34+ CD38- Using Liver Cells

Many investigators have used xenogeneic, especially murine stromal cells and fetal calf serum to maintain and expand human stem cells. The proliferation and expansion of human hematopoietic stem cells in ex vivo culture were examined with the goal of generating a suitable protocol for expanding hematopoietic stem cells for patient transplantation. Using primary fetal liver cells, we established...

Hmgb3 deficiency deregulates proliferation and differentiation of common lymphoid and myeloid progenitors.

Hmgb3 is an X-linked member of a family of chromatin-binding proteins that is expressed in primitive hematopoietic cells capable of long-term hematopoietic repopulation. To examine the role of Hmgb3 in adult hematopoiesis, we generated Hmgb3-deficient (Hmgb3(-/Y)) mice, which are viable but erythrocythemic. Hmgb3(-/Y) mice contain normal numbers of hematopoietic stem cells (HSCs), which generat...

Human Mesenchymal Stem Cells and Their, Clinical Aapplication

There are two main categories for stem cells a cording to their origin: Embryonic Stem Cells and Adult Stem Cell. Mesenchymal stem cell, supporting hematopoetic stem cells in bone marrow, can regenerate tissues such as bone, cartilage, muscle, tendon and fatty tissue. These cells were recognized for the first time by Friedenstein and Petrokova who could isolate theme from rat bone marrow.Mesenc...

EXPANSION OF HUMAN CORD BLOOD PRIMITIVE PROGENITORS IN SERUM-FREE MEDIA USING HUMAN BONE MARROW MESENCHYMAL STEM CELLS

Ex vivo expansion of human umbilical cord blood cells (HUCBC) is explored by several investigators to enhance the repopulating potential of HUCBC. The proliferation and expansion of human hematopoietic stem cells (HSC) in ex vivo culture was examined with the goal of generating a suitable clinical protocol for expanding HSC for patient transplantation. Using primary human mesenchymal stem ...

Dendritic Cells and Their Role in Cancer Immunotherapy

Dendritic cells (DCs) are antigen presenting cells with unique capability to take up and process antigens in the peripheral blood and tissues. They subsequently migrate to draining lymph nodes where they present these antigens and stimulate naive T lympho-cytes. During their life cycle, DCs go through two maturation stages and are referred to as immature and mature cells, respectively. While im...
